Overview
Seamless cloning is a simple, efficient and rapid DNA cloning technique that enables the directional cloning of DNA inserts into any site of any vector. Independent of T4 DNA ligase and free from restrictions imposed by restriction enzyme sites of vectors and target fragments, this product directly employs homologous recombination. Using a specialized enzyme mix, it directionally recombines any linearized vector with PCR fragments containing 20–25 bp overlapping regions complementary to both ends of the linearized vector. Directional seamless cloning of 1 to 10 fragments can be rapidly achieved within 5–30 minutes at 50 °C.
2×Ultra Cloning MasterMix V2 is manufactured using an optimized, state-of-the-art process, enabling highly efficient homologous recombination of 1 to 10 DNA fragments. It not only improves cloning efficiency for complex systems such as large vectors and homology arms with high or low GC content, but also enhances compatibility with variable DNA input amounts, greatly mitigating the reduction in cloning efficiency caused by low DNA fragment concentrations.
