Description
Thermostable inorganic pyrophosphatase was purified from recombinant E. coli strain, carrying the inorganic pyrophosphatase gene cloned from the extremely heat-resistant Thermococcus litoralis. Thermostable Inorganic Pyrophosphatase catalyzes the hydrolysis of inorganic pyrophosphate to form orthophosphate: P2O74-+H2O+PPase→2HPO42-. This enzyme has 100% activity when heated at 100°C for 4 h, so it can be used in PCR reaction to remove the inhibition of inorganic pyrophosphate on PCR and enhance DNA replication. Under natural conditions, PPase can provide thermodynamic power for the biosynthesis of protein, RNA and DNA, and play a role in lipid metabolism, calcium absorption and other biotransformation processes.
Main uses of this product: increase the production of transcribed RNA in vitro, increase the production of DNA in PCR reactions, remove PPi contaminants in the reagents used for SNP genotyping by pyrophosphate assay, etc.
Thermostable Inorganic Pyrophosphatase (Glycerol-Free) is a glycerol free formulation of our regular Thermostable Inorganic Pyrophosphatase and is well suited for automated high throughput testing, lyophilization or where accurate pipetting of small volumes are crucial. Thermostable Inorganic Pyrophosphatase (Glycerol-Free) is available in different sizes and concentrations.
Quality Control
After multiple column purifications, only a clear single target band was visible in SDS-PAGE gel detection; qPCR method detection showed no residual E. coli DNA and no endonuclease or exonuclease contamination.

Notes
The enzyme is active in a variety of reaction buffers. Therefore, it can be directly used in thermophilic helicase-dependent amplification (tHDA), PCR, and sequencing.
The dosage of the enzyme needs to be optimized in different experiments, usually in the concentration of 0.05-1U/ml.
