RBBP6 (Retinoblastoma-Binding Protein 6, also known as PACT or P2P-R) is a multifunctional protein that plays a central role in cell cycle regulation, transcription, and DNA damage repair. This protein contains multiple conserved domains, including an N-terminal ubiquitin-like DWNN domain, a CCHC zinc finger domain, a RING finger-like domain (possessing E3 ubiquitin ligase activity), as well as binding domains for p53 and the retinoblastoma protein (pRB). RBBP6 is a key negative regulator of the p53 signaling pathway; it promotes p53 degradation by enhancing MDM2-mediated ubiquitination, exhibiting E3 ubiquitin ligase-like activity. Concurrently, it is involved in mRNA 3'-end processing, cell cycle regulation, and chaperone-mediated protein quality control. Research indicates that RBBP6 is overexpressed in various tumors such as breast cancer and colon cancer, and its high expression is associated with poor patient prognosis. Therefore, it is regarded as a potential cancer biomarker and therapeutic target, holding significant research value particularly for tumors maintaining wild-type p53. Answer: We recommend performing a serial dilution. For example, if human serum requires a 1,000,000-fold dilution, you can use a three-step 100× dilution method as follows: Step 1: Take 2 uL of human serum and add it to 198 uL of sample diluent, mix thoroughly to obtain a 100-fold diluted sample. Step 2: Take 2 uL of the 100-fold diluted sample and add it to 198 uL of sample diluent, mix thoroughly to obtain a 10,000-fold diluted sample. Step 3: Take 2 uL of the 10,000-fold diluted sample and add it to 198 uL of sample diluent, mix thoroughly to obtain a 1,000,000-fold diluted sample. If further dilution is required, additional serial steps can be used. We do not recommend using any other solutions or water for dilution, except for the sample diluent we suggest.
