HSP47 is also named as Serpin H1, colligen, CBP1, CBP2, HSP47, SERPINH2 and belongs to the serpin family. This gene encodes a member of the serpin superfamily of serine proteinase inhibitors. The encoded protein is localized to the endoplasmic reticulum. It binds specifically to collagen and can be involved as a chaperone in the biosynthetic pathway of collagen. The Serpin Family H Member 1 (SERPINH1) gene encodes a protein called HSP47, which is required for the correct folding and secretion of collagen. SERPINH1 has been implicated in several collagen-related diseases, such as, brittle bone disease, keloid scars, and lung fibrosis. SERPINH1 also correlates with the grade of glioma, and regulates proliferation, migration and invasion of glioma cell lines. SERPINH1 regulates the Wnt/β-catenin signaling pathway and modulates gastric cancer (GC) progression SERPINH1 is aberrantly expressed in cervical cancer, breast cancer, glioblastoma and colorectal cancer. SERPINH1 promotes invasion and metastasis of breast cancer cells by regulating the expression of several extracellular matrix (ECM) proteins . Answer: We recommend performing a serial dilution. For example, if human serum requires a 1,000,000-fold dilution, you can use a three-step 100× dilution method as follows: Step 1: Take 2 uL of human serum and add it to 198 uL of sample diluent, mix thoroughly to obtain a 100-fold diluted sample. Step 2: Take 2 uL of the 100-fold diluted sample and add it to 198 uL of sample diluent, mix thoroughly to obtain a 10,000-fold diluted sample. Step 3: Take 2 uL of the 10,000-fold diluted sample and add it to 198 uL of sample diluent, mix thoroughly to obtain a 1,000,000-fold diluted sample. If further dilution is required, additional serial steps can be used. We do not recommend using any other solutions or water for dilution, except for the sample diluent we suggest.
