Nucleobindin-2 (NUCB2) is a protein encoded by the NUCB2 gene in humans. It is widely expressed in various tissues, including the brain, pancreas, and adipose tissue. NUCB2 is known for its role in energy homeostasis, appetite regulation, and glucose metabolism. The protein is a precursor for nesfatin-1, a peptide hormone that has been implicated in the regulation of food intake and body weight. Nesfatin-1 is derived from the N-terminal fragment of NUCB2 and has been shown to exert anorexigenic effects, reducing food intake and promoting satiety. NUCB2/nesfatin-1 has also been associated with other physiological processes, including stress responses, reproductive functions, and cardiovascular regulation. The protein is thought to interact with various signaling pathways, including those involving leptin and insulin, to modulate energy balance and metabolic functions. Answer: We recommend performing a serial dilution. For example, if human serum requires a 1,000,000-fold dilution, you can use a three-step 100× dilution method as follows: Step 1: Take 2 uL of human serum and add it to 198 uL of sample diluent, mix thoroughly to obtain a 100-fold diluted sample. Step 2: Take 2 uL of the 100-fold diluted sample and add it to 198 uL of sample diluent, mix thoroughly to obtain a 10,000-fold diluted sample. Step 3: Take 2 uL of the 10,000-fold diluted sample and add it to 198 uL of sample diluent, mix thoroughly to obtain a 1,000,000-fold diluted sample. If further dilution is required, additional serial steps can be used. We do not recommend using any other solutions or water for dilution, except for the sample diluent we suggest. If your sample diluent is still insufficient, please contact our customer service for more sample diluent.
