Product Description/Introduction
The presence of protein in the urine is known as proteinuria, or urinary protein, which has a more complex composition, including albumin (Alb), IgG, IgA, IgM, light chains (κ, λ), β2-microglobulin (β2M), α1-microglobulin (α1M), α2-macroglobulin (α2M), free hemoglobin (Hb), myoglobin (Mb), other plasma proteins from the plasma and enzymes, and non-plasma proteins such as Tamm-Horsfall protein (THP), glomerular basement membrane (GBM) antigen, and other derived proteins. Healthy adults can contain a small amount of protein in the urine, generally 40-80mg/24 h, if the protein content in the urine continues to be >100 mg/L or >150 mg/24 h, then the protein qualitative test is positive that is proteinuria, the increase in protein content in the urine marks the glomerular filtration membrane or tubular pathology, the renal reabsorption function decreases, it is an important indicator for early diagnosis of renal damage, and it is also an important indicator for early diagnosis of renal damage. Therefore, urine protein test is of great significance for the diagnosis and prognosis evaluation of kidney diseases, cardiovascular and cerebrovascular diseases, hypertension and other diseases.
This product is suitable for the detection of protein content in urine. Detection principle: In acidic solution, CBB combines with NH3+ group of protein to cause color reaction, showing blue color, with maximum absorption peak at 595nm, and its absorbance is proportional to the concentration of urinary protein, so it can be used for the determination of the concentration of urinary protein in the sample.

Assay Protocol / Procedures
1. Sample requirements
If the sample is clear, it can be measured directly; if it is turbid, centrifuge the sample at 10,000 g for 10 min at 4°C, and take the supernatant for detection.
2. Measurement of urine protein content
a) Standard curve production (optional)
① Standard dilution: Use the Standard Diluent to dilute the standard ( 10 g/L ) gradient into 1600, 1200, 800, 400, 200, 100, 50, 0 mg/L.
② Add 5 μL of each concentration standard to each well, detect OD595, and subtract the blank from the OD595 of each concentration standard to obtain △OD595.
③ The standard curve y=ax+b was made with the concentration of standard as the horizontal coordinate x and △OD595 as vertical coordinate y.
b) Operational steps
Mix well, incubate at room temperature for 5 min, read the absorbance value at 595 nm

Standard curve calculation：
Sample urine protein level（mg/L）=（Asample-Ablank-b）/a
Note
1. Before using the detection solution, it should be restored to room temperature and inverted to mix well, so as not to affect the sensitivity of the detection.
2. If the urine protein concentration in the sample is higher than the linear range, it can be diluted with the standard diluent and then tested again.
3. For your health and safety, please wear lab coat and gloves during operation.
