Molecular Structural
Introduction
Alcian Blue is a copper-titanium anthocyanine conjugate dye, a cationic dye that forms insoluble complexes with acidic groups, i.e., Alcian Blue, and anionic groups such as carboxylate and sulfate contained in tissues. Alcian Blue consists of a central copper-containing phthalocyanine ring with four isothiourea groups linked by a thioether bond. This isothiourea group is moderately basic, making Alcian Blue cationic. Different pH values of the stain can be used to differentiate between classes of mucus material. At pH 2.5 the carboxyl groups in the tissue ionize and carry a negative charge, forming a salt bond with the cations in Alcian Blue, which stains tissues with carboxyl groups (e.g., proteoglycans/hyaluronic acid, as well as epithelial acidic mucins). At a pH of 1.0, the sulfate in the tissue ionizes, carries a negative charge, and forms a salt bond with the cations in Alcian Blue, causing tissue with sulfate (e.g., sulfate mucus material) to stain. Neutral mucins (e.g., those in the gastric mucosa and Brunner's gland sites) do not react with Alcian Blue.
Instructions
The usage and dosage of biochemical reagents are mainly determined according to the customer's experimental purpose and the experimental methods in existing literature or books. The following uses are for reference only.
1. For bone marrow smear staining: Bone marrow smear was first fixed with methanol for 5 min, then washed and air-dried; Alcian Blue solution (10 g/L Alcian Blue aqueous solution 100 mL and 1% acetic acid 100 mL mixed and filtered) was stained for 30 min, washed thoroughly, then re-stained with 1 g/L Neutral Red aqueous solution for 30 s, washed, air-dried, and examined microscopically.
2. Alcian Blue can be used for cartilage staining, using a concentration of 0.1% Alcian Blue Staining Solution, preparation method reference: 80mL anhydrous ethanol + 20mL glacial acetic acid + 0.1g Alcian Blue8GX, fully mixed and dissolved, 37 ℃ static overnight.
Note:
1. The dyeing is too dark or too light: control the dyeing time.
2. Uneven staining of slices: ① Improper fixation of the material, standard and standard for the material, and complete fixation; ② Insufficient dehydration, re-dehydration, re-transparency, and re-wax dipping.
3. The staining of the section is blurry: ① The temperature of the embedding wax is too high; ② The dehydration and transparency are incomplete.
