Product Introduction
Swe Cell Transfection medium (Opti-MEM) is an improved basic culture medium (MEM) that contains various amino acids, vitamins, and trace elements required for cell culture. According to different cell types, the addition of fetal bovine serum can be reduced by 50 % -90 % without affecting growth rate or morphological changes.
The Swe Cell Transfection Medium can be used in conjunction with PEI 40K transfection reagent, resulting in high transfection efficiency. It has been verified that the linear PEI 40K transfection reagent is widely applicable to various cell lines including HEK-293, HEK293T, CHO-K1, COS-1, COS-7, NIH/3T3, HepG2, and Hela cells. Swe Cell Transfection Medium is also recommended for use with cationic lipid transfection reagents, such as Swe TransDNATM and Swe TransRNATM, and can be used for DNA plasmid, SiRNA and other transfection objects.
Storage and Shipping Conditions
The product is transported at room temperature, and protected from light at 2~8 °C. It is valid for two years.

Procedures
For DNA:
1. Seed cells to be 70–90 % confluent at transfection;
2. Dilute Swe TransDNATM Reagent in Swe Cell Transfection Medium;
3. Dilute DNA in Swe Cell Transfection Medium;
4. Add diluted DNA to diluted Swe TransDNATM Reagent (1:1 ratio);
5. Incubate for 5 minutes at room temperature;
6. Add DNA-lipid complex to cells;
7. Incubate cells for 1–3 days at 37 °C. Then analyze transfected cells.
For siRNA:
1. Seed cells to be 60–80 % confluent at transfection;
2. Dilute Swe TransRNATM Reagent in Swe Cell Transfection Medium;
3. Dilute SiRNA in Swe Cell Transfection Medium;
4. Add diluted SiRNA to diluted Swe TransRNATM Reagent (1:1 ratio);
5. Incubate for 5 minutes at room temperature;
6. Add SiRNA-lipid complex to cells;
7. Incubate cells for 1–3 days at 37 °C. Then analyze transfected cells.
Note
1. This product has been filtered and sterilized. When using it, attention should be paid to aseptic operation to avoid contamination;
2. Avoid freezing the culture medium, as freezing may cause nutrient precipitation and may not completely dissolve after reheating, affecting the effectiveness of cell culture. In addition, when storing the culture medium in a refrigerator at 2-8 ℃, do not store it tightly against the inner wall of the refrigerator to prevent the medium from freezing due to low local temperatures
