Product Description
This product is a powdered Ham's F-12 medium, contains 10 mM D-Glucose, 1.0 mM L-Alanyl-L-Glutamine, 1.0 mM sodium pyruvate and phenol red indicator, without HEPES. The powder forms of culture medium require sodium bicarbonate supplementation, pH adjustment, and filtration at the time of preparation (see protocols below for details).
Storage and Shipping Conditions
Ship at room temperature; Store away from light at 2-8℃, valid for 36 months.
General guidelines
1. Prepare a clean container with a volume close to the final volume.
2. Prepare distilled water at room temperature (15-30℃) or cell culture-grade injection water (recommended as G4701).
3. Prepare sodium bicarbonate, 1 N NaOH (1 mol/L) and 1 N HCl (1 mol/L).
Procotols of liquid media preparation from powder
1. Pour 95% distilled water (by final volume) into a prepared clean container.
2. Weigh the powder according to the usage ratio (10.84 g/L) and dissolve it in the water. Note that the powder will disintegrate in water and dissolve quickly. High-speed stirring might cause the powder to clump and form a water film on the surface, which is not conducive to dissolution. Therefore, there is no need for high-speed stirring after the powder is added to the water.
3. After the powder has completely dissolved, add sodium bicarbonate in a dosage of 1.176 g/L and stir to dissolve it.
4. Adjust the pH to be 0.2-0.3 units lower than the working pH (recommend a working pH of 7.0-7.4) using 1 N NaOH (1 mol/L) and 1 N HCl (1 mol/L). The pH of the solution will increase by 0.1-0.3 units after filtration.
5. Make up the volume with water to the final volume. The solution should be kept sealed before filtration.
6. Use a 0.2 μm filter membrane to filter into a sterile container (note: perform aseptic operation).
7. After filtration, take a small amount of the liquid for aseptic inspection. Use it after it is qualified. The prepared liquid medium should be stored at 2-8℃ and has a shelf life of 1 year.
Note
1. Dry powder is prone to absorbing moisture. After taking it out, seal it promptly.
2. When taking out the dry powder, take precautions to prevent inhalation.
3. During the stirring and filtration processes, keep the container holding the culture medium covered and avoid excessive stirring. Stop stirring once the dry powder has dissolved.
