Product Information
| Product Name: TTC Stain (embryo viability) | Cat. No.: G 1074-100ML |
| Spec.: 100 mL |
Product Description
Seed vigor is a key index for evaluating seed germination ability. To evaluate seed vigor correctly, germination percentage and germination potential are usually measured, which is time-consuming, laborious and complicated. TTC staining can be used to determine seed vigor simply, quickly and accurately. The dyeing principle is that there are a lot of dehydrogenase in the viable embryo, which can convert TTC (2,3,5-triphenyltetrazolium chloride) is reduced to insoluble red TTF (triphenylformazan). If the embryo dies or its vitality declines and dehydrogenase decreases, it cannot be stained or the staining will be relatively light. This TTC Stain is simple to operate, the staining is bright and the contrast is obvious. Simply soak the seeds in water and then place them in the TTC Stain to clearly and intuitively display the vitality of the embryos. Embryos with vitality appear red, while those without vitality are colorless, and weakly vital embryos show a light red color.

Assay Protocol
Sample preparation: The seeds are soaked in water at 30℃ for 2-6 h or longer to ensure they are fully imbibed.
Dyeing steps:
1. The TTC dye solution is preheated at 37℃ for 30 min.
2. The seeds are peeled from the shell or seed coat, cut longitudinally to preserve the intact embryo, and placed into a clean centrifuge tube. The samples are immersed in the TTC dye solution and stained in a water bath at 37℃ in the dark for 30–60 min. The coloring condition is observed every 15 min.
3. When the seeds are red and no longer darking, pour out the dye solution, then slowly add fixing solution (recommended G1101) to immerse the tissue to terminate the staining reaction. Protect the sample from light.
4. Remove the tissue, blot the surface with filter paper to remove excess fixing solution, and place it on a white or light-colored background plate. Adjust the camera’s focal length and take photos.
Note
1. The dyed sample fades easily, so take photos as soon as possible.
2. For your safety and health, please wear lab coat and disposable gloves.
3. Plant seed samples should be transported at room temperature, not fixed in fixative or transported at low temperature.
