General Information
| Formulation | Lyophilized from sterile PBS, pH 7.4. Please contact us for any concerns or special requirements. Normally 5 % - 8 % trehalose, mannitol and 0.01% Tween80 are added as protectants before lyophilization. Please refer to the specific buffer information in the hardcopy of datasheet or the lot-specific COA. |
| Endotoxin | < 1.0 EU per μg of the protein as determined by the LAL method |
| Shipping | In general, recombinant proteins are provided as lyophilized powder which are shipped at ambient temperature. Bulk packages of recombinant proteins are provided as frozen liquid. They are shipped out with blue ice unless customers require otherwise. |
| Stability & Storage | Samples are stable for up to twelve months from date of receipt at -20℃ to -80℃. Store it under sterile conditions at -20℃ to -80℃. It is recommended that the protein be aliquoted for optimal storage. Avoid repeated freeze-thaw cycles. |
| Reconstitution | To find reconstitution info, click on 'COA' at the top of the page, enter the lot number and product size for the lot-specific COA. |
| Purity | > 95 % as determined by SDS-PAGE |
| Activity | 1. Measured by its binding ability in a functional ELISA. Immobilized human CD226 at 2 μg/ml (100 μl/well) can bind human CD112 with a linear range of 6.4-800 ng/ml. 2. Immobilized PVRIG Protein, Cynomolgus, Rhesus, Recombinant (His Tag)(Cat:90971-C08H) at 2 μg/ml (100 μl/well) can bind Nectin-2 Protein, Human, Recombinant (hFc Tag)(Cat:10005-H02H), the EC 50 of Nectin-2 Protein, Human, Recombinant (hFc Tag)is 0.2-1.2 ng/mL. 3.Loaded Human PVRIG Protein, mFc Tag(Cat.No.28312-H38H2) on AMC Biosensor, can bind Human Nectin-2 protein, hFc Tag (Cat.No.10005-H02H) with an affinity constant of 8.784nM as determined in BLI assay (Sartorius Octet Red384) (Routinely tested). |
| Protein Construction | A DNA sequence encoding the extracellular domain (Met 1-Leu 360) of human CD112 (NP_002847.1) was fused with the Fc region of human IgG1 at the C-terminus. |
| Accession# | NP_002847.1 |
| Expressed Host | HEK293 Cells |
| Species | Human |
| Predicted N Terminal | Gln 32 |
| Molecule Mass | The recombinant mature human CD112/Fc chimera is a disulfide-linked homodimeric protein. The reduced monomer consists of 567 amino acids and has a calculated molecular mass of 62.3 kDa. As a result of glycosylation, the rh CD112/Fc monomer migrates as an approximately 70-80 kDa protein in SDS-PAGE under reducing conditions. |

Background Information
| Full Name | poliovirus receptor-related 2 (herpesvirus entry mediator B) |
| Description | Cluster of Differentiation 112 (CD112), also known as poliovirus receptor related protein 2 (PVRL2 or PRR2), is a single-pass type I transmembrane glycoprotein belonging to the Immunoglobulin superfamily. CD112 protein also serves as an entry for certain mutant strains of herpes simplex virus and pseudorabies virus, and thus is involved in cell to cell spreading of these viruses. CD112 protein has been identified as the ligand for DNAM-1 (CD226), and the interaction of CD226/CD112 protein can induce NK cell- and CD8 T cell-mediated cytotoxicity and cytokine secretion. CD112 has been regarded as a critical component in allergic reactions, and accordingly may function as a novel target for anti-allergic therapy. |
