| Description | Claudin-18.2 presents a unique challenge for recombinant production because its four-transmembrane topology demands a native membrane context to preserve conformational epitopes critical for antibody recognition. This fluorescent VLP-displayed construct presents full-length human CLDN18.2 (aa 1–261, isoform A2) with a C-terminal mCherry tag, enabling direct visualization in flow cytometry-based antibody screening and cell-based binding assays. Functional ELISA confirms that immobilized CLDN18.2 VLPs bind an anti-CLDN18.2 recombinant antibody with an EC50 of 5.1–8.5 ng/mL, demonstrating preserved extracellular loop conformation suitable for antibody development and validation, therapeutic antibody benchmarking, and tumor metastasis research targeting CLDN18.2-positive cancers. The mammalian-expressed VLP format maintains native folding and membrane orientation while endotoxin levels below 1.0 EU/μg satisfy criteria appropriate for immune cell trafficking studies and integrin-ligand binding assays requiring low background activation. |
