Overview
This kit utilizes the poly(A) tailing method for the synthesis of miRNA first-strand cDNA.
The miRNA Enzyme Mix contains E. coli Poly(A) Polymerase, RTase and RNase inhibitor.
E. coli Poly(A) Polymerase not only exhibits high poly(A) tailing efficiency but also specifically recognizes single-stranded miRNA, thereby preventing further reverse transcription of double-stranded miRNA precursors.
The 2× miRNA RT Mix in this product contains all reagents and primers required for both the miRNA tailing reaction and reverse transcription reaction. After careful optimization, it ensures efficient simultaneous execution of the 3′-end poly(A) modification and reverse transcription of miRNA.
For subsequent qPCR analysis, only a specific forward primer needs to be designed, and together with the universal reverse primer provided in the kit, miRNA in samples can be accurately detected.
