MSL2 was immunoprecipitated using: Lane A:0.5 mg Hela Whole Cell Lysate 4 µL anti-MSL2 rabbit polyclonal antibody and 15 μl of 50 % Protein G agarose. Primary antibody: Anti-MSL2 rabbit polyclonal antibody,at 1:100 dilution Secondary antibody: Dylight 800-labeled antibody to rabbit IgG (H+L), at 1:5000 dilution Developed using the odssey technique. Performed under reducing conditions. Predicted band size: 44 kDa Observed band size: 44 kDa
MSL2 was associated with those genes responding to the virus. Mechanistically, MSL2 could maintain HBV cccDNA stability through degradation of APOBEC3B by ubiquitylation in hepatoma cells. MSL2 is an E3 ligase that ubiquitylates itself and the other associated components when their stoichiometry is unbalanced, uncovering proteasome-dependent degradation as an additional layer of homeostatic control of MSL levels. MSL2, together with MSL1, has robust histone ubiquitylation activity that mainly targets nucleosomal H2B on lysine 34 (H2B K34ub), a site within a conserved basic patch on H2B tail.
