General Information
| Formulation | Supplied as sterile 50 mM Tris-HCl, pH 7.5, 50-300mM NaCl, 10mM glutathione, 0.1mM EDTA, 0.25-1.0mM DTT, 0-0.1mM PMSF, 10-25% glycerol Please contact us for any concerns or special requirements. Please refer to the specific buffer information in the hardcopy of datasheet or the lot-specific COA. |
| Shipping | Enzymes are highly recommended to be shipped at frozen temperature with dry ice. Shipment made at ambient temperature may seriously affect the activity of the ordered products. |
| Stability & Storage | Store product at –70°C. For optimal storage, aliquot target into smaller quantities after centrifugation and store at recommended temperature. For most favorable performance, avoid repeated handling and multiple freeze/thaw cycles. |

Image / Experimental Information
| Image 1 Description | Determination of ATP Km for recombinant protein kinases GLK (Cat. No.: M25-11G) using ADP-GloTM Assay. With saturating substrate concentration, the relative luminescence values were plotted against various ATP concentrations. The Michaelis-Menten equation was then used to generate the Km values. Background Information
| Description | MAP4K3 (also named germinal-center kinase-like kinase, GLK) is a serine/threonine kinase, which belongs to the mammalian Ste20-like kinase family. MAP4K3 is required for robust cell growth in response to amino acids. MAP4K3 positively regulates preterm skeletal muscle satellite cell myogenesis, but may not regulate mTORC1 activity. MAP4K3 may play a role in mTORC1 full activation in response to leucine. MAP4K3 is required for phosphorylation of known mTOR targets such as S6K1 (S6 kinase 1), and overexpression of MAP4K3 promotes the rapamycin-sensitive phosphorylation of these same targets.MAP4K3 is a new component in the nutrient-responsive pathway for mTOR activation and reveals a completely new function for MAP4K3 in promoting cell growth. |
